Androgen receptor in nuclei of rat testis.

Testis nuclei of hypophysectomized rats selectively accumulate labeled testosterone and 5alpha-dihydrotestosterone following the injection of tritiated testosterone in vivo. Testosterone and 5alpha-dihydrotestosterone are bound to macromolecules in nuclei and can be extracted with 0.5 M KCl. Accumulation of protein bound radioactive androgens in nuclei of isolated seminiferous tubules is similar to that of whole testis. The relative amounts of testosterone and dihydrotestosterone in purified nuclei were similar to the relative amounts bound to cytoplasmic receptors, suggesting that cytoplasmic androgen-receptor complexes may be transported into the nuclei. Binding of labeled androgen is saturable and inhibited by prior injection of unlabeled testosterone or cyproterone acetate. Nuclear binding sites are destroyed by the proteolytic enzyme pronase, but not by DNase. Like the cytoplasmic androgen-receptor complexes in rat testis, nuclear androgen-protein complexes are heat labile and dissociate slowly at 0 degrees C. androgens fail to accumulate in testis nuclei of the Stanley-Gumbreck androgen insensitive rat, a species lacking cytoplasmic androgen receptors in testis and other androgen target tissues.     

Translocation (13q21q)

Summary A (13q21q) translocation was found in an infant with Down's syndrome. The 17-year-old mother and the grandmother carried the translocation 45,XX,t(13;21)(p12;q11). The great grandparents had normal karyotypes. Fluorescence marker studies suggested that the translocation originated in the great grandmother. The hypothesis was supported by satellite association studies which showed a significant excess of 13–21 and 13–15 associations in the great grandmother.     

Alterations in the lipid content of rat palatal epithelium during carcinogenesis

Summary The hard palates of 150 female albino rats of the Sprague-Dawley strain were painted 3 times a week either with the fat-soluble carcinogen 7,12-dimethylbenz(a)anthracene (DMBA) for 11 months with inhibited secretion of saliva or with the water-soluble carcinogen 4-nitrochinoline N-oxide (4NQO) for 8 months with intact salivary secretion. Specimens were taken regularly from the mucosa of the hard palate, and the content of lipids in the epithelium was studied histochemically and biochemically during the carcinogenesis.Changes in the lipid content could be observed histochemically as there was a focal loss of lipid stainability in the epithelium during the more advanced stages of carcinogenesis with severe epithelial dysplasia, carcinoma in situ and early invasive carcinoma.The biochemical method used (TLC) could not, however, verify the histologically observed changes in the lipid content of the epithelium, probably because the changes were very local.The investigations have been performed with financial support from Maggie Stephens Foundation, Colgate-Palmolive Limited and from the Swedish Dental Society.     

Controlled study of atenolol in treatment of hypertension.

The antihypertensive effect of atenolol, a new beta-1-receptor blocking agent, was studied in a double-blind trial in which 45 patients with essential hypertension were randomly assigned to placebo or atenolol treatment. Atenolol caused a statistically significant and clinically relevant reduction of blood pressure. The optimum daily dose for moderately severe hypertension was considered to be 200 mg. Several irrelevant side effects were collected by the use of a check list, but there was no difference in the number of complaints during placebo and active treatment. Atenolol has a useful antihypertensive effect and, at least theoretically, has advantages over other beta-adrenergic blocking agents.     

No Diagnostic Value of Plasma Clusterin in Alzheimer's Disease

There is an urgent need for biomarkers to enable early diagnosis of Alzheimer''s disease (AD). It has recently been shown that a variant within the clusterin gene is associated with increased risk of AD and plasma levels of clusterin have been found to be associated with the risk of AD. We, therefore, investigated the diagnostic value of clusterin by quantifying clusterin using an ELISA in plasma from 171 controls, 127 patients with AD, 82 patients with other dementias and 30 patients with depression. We observed similar plasma clusterin levels in controls, AD patients and patients with other dementias, suggesting that plasma clusterin levels have no diagnostic value for AD. There was a slight, but significant, increase in plasma clusterin in patients with depression compared to all other groups tested, which may warrant further investigation.
